چکیده

Evidence that the glutamine synthetase dodecamer from Escherichia coli is comprised of 12 identical subunits of molecular weight 49,000 has been obtained by end group analysis. Application of the carbamylation procedure to the NH*terminal analysis revealed only serine. The yield was 98% of that expected from a polypeptide of the size proposed for the monomeric species. The following partial sequence of residues at the NH2 terminus of the molecule was elucidated with the aid of automated sequencing equipment:

Maize glutamine synthetase cDNAs: isolation by direct genetic selection in Escherichia coli.

Maize glutamine synthetase cDNA clones were isolated by genetic selection for functional rescue of an Escherichia coli delta glnA mutant growing on medium lacking glutamine. The Black Mexican Sweet cDNA library used in this study was constructed in pUC13 such that cDNA sense strands were transcribed under the control of the lac promoter. E. coli delta glnA cells were transformed with cDNA libra...

Identification of a reactive cysteine residue at the glutamine binding site of carbamyl phosphate synthetase.

Carbamyl phosphate synthetase from Escherichia coli, which is composed of a light and a heavy subunit, is inhibited with respect to its glutamine-dependent reactions by treatment with ~-2-amino-4-oxo-5-chloro[5-‘~C]pentanoic acid. This glutamine analog reacts with a glutamineor albizziinprotectable site on the light subunit to yield an enzyme covalently linked to a 4-oxo[W]norvaline moiety. The...

The amino acid sequence in the vicinity of the covalently bound adenylic acid in glutamine synthetase from Escherichia coli.

Under certain conditions of growth, the Escherichiu coli glutamine synthetase is modified by the introduction into the enzyme molecule of covalently bound adenylic acid. This mechanism for modification of the preformed enzyme was discovered independently in this country by Kingdon and Stadtman (1, 2) and in Germany by Holzer et al. (3, 4). An enzyme was described by Kingdon, Shapiro, and Stadtm...

Cloning, Expression, and Purification of Glutamine Synthetase from Clostridium acetobutylicum.

A glutamine synthetase (GS) gene, glnA, from the gram-positive obligate anaerobe Clostridium acetobutylicum was cloned on recombinant plasmid pHZ200 and enabled Escherichia coli glnA deletion mutants to utilize (NH(4))(2)SO(4) as a sole source of nitrogen. The cloned C. acetobutylicum gene was expressed from a regulatory region contained within the cloned DNA fragment. glnA expression was subje...

Amino acid sequence of Escherichia coli glutamine synthetase deduced from the DNA nucleotide sequence.

Glutamine synthetase is encoded by the glnA gene of Escherichia coli and catalyzes the formation of glutamine from ATP, glutamate, and ammonia. A 1922-base pair fragment from a cDNA containing the glnA structural gene for E. coli glutamine synthetase has been sequenced. An open reading frame of 1404 base pairs encodes a protein of 468 amino acid residues with a calculated molecular weight of 51...
